Abstract:
Protein-protein interactions determine the outcome of most cellular processes. We developed a reverse two-hybrid system for the identification of critical residues for protein-protein binding and applied this method to the characterization of the autophagic machinery in the yeast model. We focused on the evolutionary conserved PROPPIN/WIPI proteins, which play a key role in the recruitment of different components of this machinery to the preautophagosomal structure and are associated with rare neurodegenerative diseases in human. Our results reveal the structural basis of the functional specificity of WIPI proteins at different steps of the autophagy process.
